Quantification and modeling of clonal dynamics in hematopoiesis provide mechanistic insights into how blood development is organized and potentially disturbed in disease situations. There is a range of quantitative methods to generate and analyze clonal data in experimental and clinical settings, and to better understand treatment dynamics and to optimize therapeutic strategies. I will discuss several such approaches, focusing on clonal barcoding and clone size quantification in gene therapy trials. Additionally, I will highlight the potential of mathematical models for the treatment of leukemia and for CAR-T cell therapies, with a particular focus on tumor-immune interactions. The integration of data analysis with mathematical models holds significant promise for advancing treatments for blood disorders and improving personalized medicine strategies.
